Influence of intraventricular atropine on the centrally induced renal action of acetylcholine and 5-hydroxytryptamine in the rabbit

Abstract
As it has been shown that acetylcholine(Ach) elicits antidiuresis while 5-hydroxytryptamine(5-HT) induces diuresis in the rabbit when, they are given intracerebroventricularly(ivt. ), it was attempted in this study to clarify further the mechanism by which these agents affect the center and possible roles of these -substances in the brain function by means of atropine, a selective anticholinergic Agent.
Atropine when given ivt. in a dose of 0.4 mg did not markedly affect the renal function. The excretory rate of electrolytes tended to slightly increase, -whereas urine flow rate and renal plasma flow tended to decline.
Ach, 10 pg, ivt. , produced antidiuresis and decreased excretion of electrolytes, Concomitant with the declines in the glomerular filtration rate and renal plasma flow. These declines of various parameters of renal function by intraventricular Ach were completely abolished by the preceding administration of atropine into the lateral ventricle.
5-HT, 50 ,ug, ivt. , elicited increased urinary output of electrolytes and water along with increased renal plasma flow and glomerular filtration. After pretreatment with atropine, however, the increased output of electrolytes and water persisted, while the homodynamic effect was abolished, indicating that a part of the 5-HT action on the brain is mediated by the atropine-sensitive cholinergic receptors.
It is suggested from these observations that Ach stimulates through the atropine-sensitive cholinoceptive receptors a certain structure in the brain to increase the sympathetic influence to the kidney, and that 5-HT affects partly via atropinesensitive cholinergic mechanism. the parasympathetic center to result in increased perfusion and filtration in the kidney.
